High efficiency SPRI (Solid Phase Reverse Immobilization Paramagnetic Bead) technology for microRNA applications

Abstract
Micro RNAs (miRNAs) are small, naturally-occurring non-coding ribonucleic acids with sizes between 18 and 40 nucleotides (nts) that have been demonstrated to play a significant role in the regulation of gene expression.  As a result, interest in smaller RNA species such as miRNA has increased. This webinar describes the method development for miRNA purification, enrichment and RNA fractionation using Beckman Coulter’s SPRI (Solid Phase Reverse Immobilization) magnetic bead based chemistry and the Biomek automated extraction method.  Total RNA and miRNA can be purified from very small amounts of highly degraded FFPE (Formalin Fixed, Paraffin Embedded) and LCM (Laser Captured Microdissection) samples. The SPRI chemistry produced superior results to a column purification method for miRNA purification, miRNA enrichment and RNA size selection.  The SPRI chemistry is an easy, high-yielding and automation-friendly nucleic acid purification process that does not require centrifugation and vacuum filtration steps. Purified nucleic acids are easily eluted from the magnetic beads under aqueous conditions, which provide maximum flexibility for downstream applications.
