Optimizing mRNA Transfection of Human T-cells

Optimizing gene-delivery strategies for T-cells is crucial to enhance their therapeutic effectiveness.

The study highlighted in this application note focused on improving the transfection of human T-cells with mRNA using the jetMESSENGER® transfection reagent. The study's specific goals included identifying culture conditions that maximize cell proliferation, transfection efficiency, and cytotoxic activity against cancer cells. To aid T-cell recovery and expansion, the cells were cultured in specialized GMP media, enriched with high-quality cytokines from Sartorius. The Incucyte® Live-Cell Analysis System and the iQue® High-Throughput Screening (HTS) cytometer were employed for kinetic visualization and quantitative assessments, which guided the optimization process.
