The purification of proteins and antibodies is a critical process in biotechnology and pharmaceutical industries. This webinar discusses the development and applications of reusable Thermo Fisher Scientific supports for protein and antibody purifications, with a focus on manual and automated workflows using high-capacity Ni-IMAC (Nickel Immobilized Metal Affinity Chromatography) and Protein A resins and magnetic beads. Ni-IMAC and Protein A supports are widely utilized in protein purification processes due to their high specificity and efficiency. Ni-IMAC is particularly adept at purifying histidine-tagged proteins, leveraging the strong affinity between nickel ions and histidine residues. Protein A, on the other hand, is specialized for the purification of antibodies, exploiting the high affinity of Protein A for the Fc region of immunoglobulins. Both supports offer robust regeneration capabilities, allowing for multiple cycles of use without significant loss of binding capacity using standardized CIP (Clean in Place) protocols. The versatility of Ni-IMAC extends to various sample types including cell lysates and culture supernatants, whereas Protein A is predominantly used for antibody purification from complex biological matrices such as serum, plasma, and cell culture media.
