Histone modification is an epigenetic mechanism that enables fine-tuning of gene expression. Mutation or dysregulation of proteins associated with histone modification (called “writers,” “erasers,” and “readers”) alters normal gene expression and has been found in a variety of diseases.

Chemical modulators such as small molecule inhibitors and proteolysis-targeting chimeras (PROTACs) have been developed to study the functions of writers, erasers, and readers of histone modifications.
