Histone modification is an epigenetic mechanism that enables fine-tuning of gene expression. Mutation or dysregulation of proteins associated with histone modification (called “writers,” “erasers,” and “readers”) alters normal gene expression and has been found in a variety of diseases.

Chemical modulators such as small molecule inhibitors and proteolysis-targeting chimeras (PROTACs) have been developed to study the functions of writers, erasers, and readers of histone modifications. It is important to understand the mechanisms and characteristics of these different modulators in order to choose the most appropriate one for your application.
