Join this webinar to learn how the Bioscience Core Lab at KAUST has automated their PacBio Kinnex library prep workflow on Opentrons robots to facilitate high-throughput RNA sequencing. Angel Angelov will present an adaptable workflow to automate the Kinnex PCR step for the PacBio Kinnex full-length RNA, single-cell RNA, and 16S rRNA protocols. The workflow includes a Shiny web app where the user can modify various protocol parameters and a python protocol template to execute the protocol on the OT-2 robot. With the release of the newest Opentrons app these parameters can also be modified at runtime. Sequencing data from example Kinnex 16S rRNA and full-length RNA runs as well as points for troubleshooting the protocols will be presented.
The PacBio Kinnex kits utilize the MAS-Seq method, which concatenates smaller amplicons into larger fragment libraries for throughput increase. Kinnex kits can be used for full-length RNA, single-cell RNA, and 16S rRNA sequencing on PacBio long-read sequencing systems to enable large-scale studies at a resolution difficult to attain with short-read RNA sequencing.
Opentrons lab robots support automation of techniques important for genomics applications, such as nucleic acid extraction and NGS library prep. With modular pipettes, and on-deck modules, including a Thermocycler and Magnetic Block, users can fully automate their library prep workflows.
