The promise of tissue clearing techniques in combination with 3D-immunofluorescence (IF) microscopy is revolutionary. Complex biological systems could suddenly become available in detail, without the need for the tissue sectioning necessitated by today’s “gold standard” immunohistochemistry. You can analyze tumor-infiltrating immune cells, the architecture of rodent nervous systems, and much more, in full spatial detail without laborious processing of individual sections and the potential loss of critical information this inevitably entails. If only reliable tissue clearing and immunolabeling protocols, not to mention the “small” detail of validating antibodies for 3D-IF applications, wasn’t prohibitively time-consuming and costly. To support researchers in their endeavors, Miltenyi Biotec has developed solutions for the main steps of the cleared tissue imaging workflow.
In this talk, Markus Habich will give insight into the basics of tissue clearing and immunolabeling techniques as well as on the different challenges of tissue clearing and immunolabeling. Also, Markus will present how the use of 3D-IF-validated recombinant antibodies, easy-to-use tissue clearing reagents and protocols, and the newly developed UltraMicroscope Blaze™ Light Sheet System help to overcome these challenges.
